چکیده

1. A competitive protein-binding assay for plasma 25-hydroxycholecalciferol was developed, using Sephadex LH-20 columns for the isolation of the sterol and a partially purified protein from rat serum as the binding protein. 2. The mean plasma 25-hydroxycholecalciferol in eighteen male and female normal volunteers was 38 5 SD 14 pmol/ml (1 5.2 & 5.6 nglml). 3. Levels of 25-hydroxycholecalciferol in plasma of four male volunteers maintained for 2 months on a diet supplemented with 26-1 nmol (10 pg) of cholecalciferol a day were found to be significantly higher. 4. High correlation was established between plasma 25-hydroxycholecalciferol values obtained by the competitive protein-binding assay and values obtained by bioassay.

The absence of 24,25-dihydroxycholecalciferol in anephric patients.

1. In subjects with normal renal function there was a strong positive correlation between serum concentrations of 25-hydroxycholecalciferol and 24,25-dihydroxycholecalciferol, as measured by competitive protein-binding assay. 2. The 24,25-dihydroxycholecalciferol concentration was about 7% of the prevailing 25-hydroxycholecalciferol concentration. 3. In contrast, sera from anephric patients con...

Structural Requirements for the Interaction of lcu,25-(OF&Vitamin D, with Its Chick Intestinal Receptor System*

The mechanism by which lac,25-dihydroxycholecalciferol (lcu,25-(OH),D,), the biologically active metabolite of cholecalciferol (vitamin D,), stimulates intestinal calcium absorption has been shown to involve an interaction of the steroid with a specific cytosol-chromatin receptor system in this target organ. Thus, la,25(OH),D, binds to a specific cytoplasmic receptor protein and then, following...

Plasma levels and therapeutic effect of 25-hydroxycholecalciferol in epileptic patients taking anticonvulsant drugs.

Plasma levels of 25-hydroxycholecalciferol (25-HCC) were measured by a specific competitive protein-binding assay. Mean levels in both normal London adults and adolescent schoolchildren were 16 ng/ml and the mean level in a group of epileptic patients on high-dosage anticonvulsant therapy was 5 ng/ml, (difference from normals P < 0.001). Two further epileptic patients, with well-marked anticonv...

Optimum Conditions of Radioligand Receptor Binding Assay of Ligands of Benzodiazepine Receptors

To obtain drugs which are more selective at benzodiazepine (BZD) receptors, design and synthesis of functionally selective ligands for BZD receptors is the current strategy of our pharmaceutical chemistry department. The affinity of newly synthesized ligands is assessed by radioligand receptor binding assays. Based on our previous studies, 2-phenyl-5-oxo-7-methyl-1,3,4-oxadiazolo[a,2,3]-pyrimid...
